109: 293 Mar. ; , 1959. The rat demonstrates spontaneous coronary lesions with age, consisting usually of lipomatous intimal lesions or diffuse fibrosis and hyperplasia. In these experiments a Moenckeberg type of medial sclerosis as well as calcification, splitting of the internal elastic membrane and pronounced degrees of proliferation and lipid accumulation in the subintimal layer were noted. Frequently thrombosis and occlusion of the arterial lumen were seen in the latter. Cardiac infarction was distinguished from cardiac focal necrosis by a wall of leukocytes surrounding the necrotic tissue. The rats used in these experiments were either younger than 2 years at the time of sacrifice or died before reaching this age, so that spontaneous changes were rarely seen. Cholesterol feeding alone induced only lipomatous changes, probably due to the ability of the rat to metabolize exogenous cholesterol efficiently. Exposure to cold only increased lipomatous changes. The incidence of coronary lesions was not affected by adrenalectomy. The addition of viosterol to the cholesterol diet produced a Moenckeberg type of medial sclerosis with death usually due to nephrocalcinosis. When thiouracil was added to the above regime, 64 per cent had severe athero and emend.
QLT has lost two separate patent litigation suits with TAP Pharmaceuticals over the prostate cancer drug Eligard and MEEI over the rights to Visudyne. The damages related to the MEEI case could represent US million if applied retroactively, and those related to the TAP case could be substantially more. The damages for both cases should be settled in the early part of 2007 and could potentially have a negative impact on the company's cash reserves.
Could reflect small amounts of intact u-hCG or immunoreactive fragments persisting in the circulation, but the bioactivity of this material is unknown. The more rapid disappearance of immunoreactive r-hCG correlates with the lower antigenicity and decreased antibody formation observed with r-hCG compared to u-hCG and indicates that recombinant preparations of hCG should be of great value in nonhuman primate research. Both u-hCG and r-hCG stimulated steroidogenesis with similar efficiency. No differences were detected between these two preparations in their ability to enhance luteal P production. Estradiol levels were also increased during CG administration, as noted in previous studies 14 ; . Treatment with u-hCG or r-hCG delayed mensesto days 22-23, reflecting the elevated serum P concentrations that persisted throughout the period of CG treatment. Taken together, these data suggest that u-hCG and r-hCG were equally effective in prolonging the functional lifespan of the primate corpus luteum. The present study shows a delayed response to both u-hCG and r-hCG, with increased P detected 2-3 days after the initial injection. These results are similar to those of a previous study using u-hCG Profasi, Serono ; to simulate early pregnancy, where a 2- to 3-day lag between initial hCG administration and increased serum P levels was also reported 4 ; . Notably, serum hCG immunoreactivity was not reliably detected in the current study until 2 days after the initial CG administration. This contrasts with an earlier report 14 ; , in which enhanced levels of P and immunoreactive hCG were detected 9 h after the first u-hCG APL, Ayerst ; administration. Divergence between the results of the current study and previous work may reflect differences in the bioactivity of the hCG administered, which would be most pronounced during the initial days of treatment. The overall pattern of circulating P observed in the current study resembled that seen previously 14 ; , with P peaking before administration of the final hCG doses in the series. RLX concentrations typically did not differ between the two groups during and after treatment, indicating that u-hCG and r-hCG also effectively stimulated the peptidergic function of the corpus luteum during simulated early pregnancy. In the current study RLX levels increased 1 day after the initial hCG administration. These data are consistent with the detection of increased serum RLX concentrations in women by day 11 of the luteal phasein conceptive cycles 22 ; . An earlier study in monkeys reported a delayed responseof RLX to CG treatment, with elevated serum RLX levels first detected after 7 days of u-hCG administration 7 ; . Results for these patients are reported separately later. The primary analysis of the induction portion of the study is based on the remaining 665 patients, of whom 493 and 172 were randomized to SDAC + DNR and HDAC 2 g m' ; DNR, respectively. Characteristics of these 665 patients are summarized in Table 1. The two arms are similar with respect to the characteristics shown in Table 1, with one exception: the SDAC + DNR arm has relatively more patients younger than 50 59% ; than does arm 2 49% ; Table 2 ; . This is a consequence of the fact that HDAC was redefined as 2 g for patients younger than 50 after the study was open for approximately 2 years; patients in this age group randomized to the 3 g m' dose of HDAC before this redefinition are described separately later. Hyperleukocytosis WBC count 100, 000 pL ; was present in 13%of SDAC + DNR patients and 12% of HDAC 2 g m2 ; DNR patients. Two patients were classified by central pathology review as having MOM5 and MOA47 morphology, respectively, and 20 other patients had AML that could not be subclassified according to FrenchAmerican-British FAB ; criteria; these 22 patients are identified as "other myeloid" in Table 1. One patient on the HDAC 2 g m * ; DNR arm refused to receive protocol induction therapy, and another 17 12 SDAC + DNR and five HDAC [2 g m2] + DNR ; chose to remove themselves from treatment early. Thirty-two patients 13 SDAC + DNR and 19 HDAC [2 &m2] + DNR ; were removed from induction therapy early because of toxicity, most frequently hemorrhage, hepatotoxicity, or neurotoxicity. Toxicity. Induction toxicity could not be evaluated for five patients due to major treatment deviations, refusal to receive therapy, early death, or lack of documentation. ABSTRACT Progesterone P ; -induced PRL secretion in estradiol -primed monkeys is not due to direct pituitary stimulation, because lactotropes do not express progestin receptors PR ; . However, the hypothalamus, particularly the tuberoinfundibular dopaminergic system TIDA ; , plays a major role in the regulation of PRL secretion. To determine whether hypothalamic dopamine neurons are progestin target cells, the colocalization of PR and tyrosine hydroxylase TH ; , a phenotypic marker of dopaminergic neurons, was examined with double immunocytochemistry. Two methods for visualizing the antigens were applied; the first was a dual peroxidase method, and the second was a peroxidase-alkaline phosphatase method. In addition, the question of whether E induces PR in dopamine neurons was explored. Spayed female monkeys were treated with empty Silastic capsules, E-filled capsules for a period of 28 days, or E capsules supplemented with P capsules for the last 14 days of E treatment. Only the E- plus P-treated monkeys exhibited an increase in serum PRL during the P treatment period. Frontal sections at the level of the optic chiasm and arcuate nucleus were examined for the colocalization of TH and PR. After E treatment, hypothalamic PRpositive cells increased in both intensity and number. Neurons expressing both TH and PR were detected in the rostra1 hypothalamus, lateral to the third ventricle All-rostral ; and in a discrete subventricular population All-subvent ; . The lateral population continued caudally All-caudal ; . The All-subvent population exhibited little steroid regulation. Of the remaining All TH neurons, approximately 20% exhibited PR in the spayed and E-treated groups. Addition of P doubled the percentage of PR-containing TH neurons in this group. Although very few TH-positive neurons in the ventral arcuate nucleus contained PR AlB-ventral ; , many double labeled neurons were observed in the dorsal arcuate region A12-dorsal ; . Ventral arcuate TIDA neurons were not regulated by steroids, but E plus P increased PR expression in A12dorsal. Double labeled cells were rarely seen in the zona incerta A13 ; or the emerging ventral tegmental area AlO ; . In summary, P probably does not act directly on ventral arcuate TIDA neurons to stimulate PRL secretion. However, the frequency of PR-positive dopamine neurons in the All-rostral, All-caudal, and A12-dorsal groups increased with E and P treatment. Despite the lack of U.S. regulatory approval, in April 2004 the Centers for Medicare & Medicaid Services, or CMS, in the U.S. implemented its decision to provide coverage for Visudyne treatment to patients with AMD who have occult and minimally classic lesions that are four disc areas or less in size and show evidence of recent disease progression. Although Novartis Ophthalmics is not permitted to promote Visudyne in these indications in the U.S., sales are made in the U.S. to physicians who are treating patients with occult and minimally classic lesions. Eligard 3.75-mg one-month Product Through our licensee, Sosei, we are developing Eligard in Japan for the treatment of prostate cancer. Sosei previously submitted the application for marketing approval of the Eligard 3.75 mg one-month formulation in Japan. After interaction with the Japanese Regulatory Authorities, our partner Sosei decided to withdraw the Eligard 3.75 mg one-month dossier from the Generic Division. Sosei plans to refile the dossier with the division responsible for new formulations upon completion of additional non-clinical studies. Lemuteporfin Lemuteporfin is a proprietary photosensitizer to which we own or exclusively license all rights. We are currently developing an injectable formulation of lemuteporfin for the treatment of benign prostatic hyperplasia, or BPH. BPH is the most common prostatic disease. According to the U.S. National Institute of Diabetes and Digestive and Kidney Diseases, over 50% of men in their sixties and older have symptoms of BPH. It is a progressive condition that results from an excessive benign growth of prostatic tissue. The majority of patients with this disease will experience developing symptoms of urinary obstruction lower urinary tract symptoms ; of progressive severity. The methods of managing BPH symptoms depend upon the severity of the symptoms. Treatments range in severity from pharmacological treatment to minimally invasive therapy, and finally prostate resection. We completed enrolment of a Phase II light dose-ranging study of Lemuteporfin in BPH during the last quarter of 2005. This controlled trial enrolled 195 patients across 25 centers. In February 2006, we announced that the Phase II clinical trial of Lemuteporfin-injectable in patients with BPH did not meet the study's primary efficacy objective at three months. While the decrease in AUA American Urological Association ; Symptom Score was consistent with that seen after other minimally invasive therapies, there was no significant difference between treatment and sham-control groups. We will continue to follow-up patients that participated in that trial to determine whether there is any treatment benefit at the 6 months time point after treatment. After we have evaluated the six month data we will determine whether the data is sufficient to permit us to license this program to a third party for further development. AczoneTM Aczone is under development for the treatment of Rosacea. We initiated a Phase II study in November 2005. The enrolment is now complete and it is expected that the 12-week follow-up results will be available in 2006. Rosacea is a chronic skin disorder that most often affects the central face including the nose, forehead, cheeks and chin. Superficial small vessels become dilated, resulting in blotchy red areas with small papules and pustules and telangiectasia. Rosacea usually affects fair-skinned people 30 to 50 years of age who tend to blush or flush easily. Symptoms are complex and include flushing, erythema, papules and pustules, telangiectasia and burning stinging. Octreotide Atrigel- three-month Product We have developed a formulation of octreotide in the Atrigel delivery system for the treatment of the symptoms of acromegaly. In the first quarter of 2006, we filed an IND for this study and expect to initiate Phase IIa clinical studies in 2006 in acromegalic patients.
