VAN THOR, University of Oxford -- Distinctive photodynamic properties of the green fluorescent protein GFP ; from the jellyfish A. victoria result from charge transfer processes involving the autocatalytically generated chromophore. We investigate structural changes in response to chromophore photoionization at cryogenic temperatures, using both X-ray crystallography and polarized infrared measurements on oriented single crystals. These measurements identify conformational changes of Gln 69, Cys 70, and an associated H-bonded cluster of internal water molecules in the chromophore environment. These structural changes take place at 100 K, far below the "dynamical transition" traditionally regarded as enabling functional protein motions. This contrasts with the prevailing view that the rigid interior of the GFP -barrel sterically inhibits nonradiative processes. Instead, we propose that rapid rearrangements of the chromophore environment enhance the fluorescence quantum yield by stabilizing the abruptly altered charge distribution in the radiative state. We suggest that the conformational response to charge transfer influences two fundamental and useful spectroscopic properties of GFP--the large frequency separation between excitation and emission and the efficient fluorescence and ubiquinone. Nonneoplastic pathology. There were no gross pathologic changes in mice euthanized for plasma drug concentration analysis at 52 weeks. After 104 weeks, enlargement of interscapular brown fat was observed in males at 400 and 800 mg kg and in females at all doses. Lipid vacuoles in the brown fat increased in size and coalesced. Liver weight increased 18% in males at 400 and 800 mg kg, and 33 and 55% in females at 400 and 800 mg kg, respectively. Males given 400 mg kg and females given 800 mg kg had hepatocellular vacuolation. Heart weight increased 17 and 22% in males and 24 and 29% in females at 400 and 800 mg kg, respectively. No histopathologic findings correlated with increased heart weight. The incidence of fatty change and hypocellularity in bone marrow was increased in both sexes at all doses. Decreased kidney weights 12% ; and a decreased incidence of and triptorelin.
